[d-Arg7, d-Lys15, d-Lys23]-esculentin-2CHa(1-30) is a synthetic peptide analogue of esculentin-2CHa(1-30), an antimicrobial peptide (AMP) originally isolated from the skin secretions of the edible frog *Rana esculenta* (Pelophylax esculentus). This specific analogue incorporates D-amino acid substitutions at positions 7 (Arginine), 15 (Lysine), and 23 (Lysine) to enhance metabolic stability against proteolytic degradation. In preclinical research, the peptide has demonstrated significant therapeutic potential for the treatment of type 2 diabetes and obesity. It has been shown to reduce body weight and fat mass, lower blood glucose and HbA1c levels, and improve insulin sensitivity and glucose tolerance. Its mechanism of action involves preserving and enhancing pancreatic islet function, promoting beta-cell proliferation, and modulating the expression of key metabolic genes in skeletal muscle and pancreatic tissues, such as GLUT4, IRS1, and PDX1.
